Referat av foredraget til Prof. De Meirleir om XMRV/MLV 22.09

Because I’m tired it’s difficult to give you an impression. But I will give it a try. I want to apologize in advance for al the vocabulary mistakes I make in writing and translating.

DML gave a short presentation with the latest news on XMRV, data from the Workshop in Washington. Data we all know by now, because this forum is faster than the light. More interesting is ofcourse his personal feeling about what happened over there and therefor I will quote him on several subjects.

He mentioned the Lombardi study and the fact that in less then one month there were some negative studies published.
«It’s impossible to do good research in less then a month».

About the Blomberg study from Sweden, where they couldn’t find any XMRV, he said the following;
«We delivered 132 bloodsamples and Blomberg did find two XMRV+, but he forgot to mention them in his study. Speaking about politics».

The patiënts chosen for the Ampligenstudy were badly chosen. (33%XMRV+)

Blood from relatives tested 50% XMRV+
Lots of patiënts have children with autism

«There is a lot more to come», he stated seriously

He told us that in a personal communication with Cheney, Cheney said that he had good results in treating the patiënts with Artusenate, a NFKappaB inhibitor.

He couldn’t tell us anything about the other studies concerning the relation between XMRV and other disseases, but he confirmed ten diseases already found evidence (XMRV+ patients)

He talked about Singh and the 4 medications that has proven their effectivity. Ragrevir, AZT, Tenofovir and Zidovodine. » This combination is not tried in patiënts yet, but will possibly dangereous in regard to resistancy».

«The virus is stable. Because of the problems with the viral load that seems to be shifting anti-retrovirals are not usefull». (Here I actually nodded my head and nearly fell of my chair)

The Alter/Lo study is being replicated in Belgium and for the now he could tell us that 50% tested MLV+)

He stressed the importance of the journalist/patiëntgroup at the workshop putting pressure on the virologists for clinical trials. (personally I find that paradoxical with his earlier statement that anti-retrovirals are not usefull, nevertheless I’m philosopher and a critical mind)

DML thinks that the symptoms play a bigger role in the disease than the virus at a given moment.
When I ask him if al these treatments to get rid of the symptoms (leaky gut etc..) aren’t a waste because the virus self remains in place and active, he answers that it is a cirkle. (Personally I don’t believe that)

«They have asked me not to show the data yet, but yes it is. The study will be published soon!»

    Ja denne så jeg på i natt! Jeg vet faktisk ikke helt hva jeg skal tru på eller ikke. For å si det sånn; veldig spent på hva rapporten til KDM kommer med. Så sitter vel litt med en «vent å se holdning». Men det er klart at i forhold til den svanske studien, så kan det tyde på at de har holdt tilbake info om XMRV positive. Vi vet også at det er flere svensker som går til behandling hos både KDM og LHK. Hvor mange er uklart.

    Det vi riktignok vet og har fått forståelse for, er at det er uenighet i fagmiljøene, med ulike hypoteser. Teorier kan man kanskje ikke helt snakke om enda, selv om det foreligger mye testresultater fra både behandlere og forskingsrapporter av denne tilstanden.

    Men det er nu engang sånn, at innefor vitenskap, skal hypoteser bevises. På den andre siden er det mye forskning som er på god gli.

    …men skal jeg snakke ut i fra mine erfaringer, uten å gå i dybden, så er det maktkamp og også ett kjent problem at det tilbakeholdes informasjoner. Proaganda og manipulasjon.

    Det hviler vel heller ingen tvil om at dette er et potensielt betent område, mest fordi at det vil komme til at denne gruppen ikke har fått behandling og blitt trudd på. Amerikanere er jo kjent for sine søksmål, og jeg trur ikke at vi skal ignorere dette på hjemmebane heller – når alt kommer til alt. Erstaningskravene vil komme, og det kommer til å koste dyrt.

    Rett skal være rett, og rettferdighet må seire – nok er nok….

    Denne kampen har så vidt begynt!

    Blomberg är mycket seriös. Han är helt kommittad till att fortsätta reda ut virusfrågan. Han när också stora sympatier för de ME-sjuka. Att antyda att Blomberg har en annan agenda är bullshit.

      Sitter du med noen informasjon om XMRV-positive testet andre steder, Redlabs eller LHK (Norge) har innrapportert sine funn til Blomberg og/eller om han har samarbeid med retro-virologer/sendt prøver til USA…..

      I så tilfellet betviler jeg ikke Blombergs seriøsitet!

    Jag tror det brister lite i dina kunskaper. Själva poängen med ett forskningsfynd är att det måste gå att upprepa. Det är därför oberoende lab försöker göra om det som WPI nu grundlagt. När det gäller MLV-virus så hade Blomberg en föreläsning på XMRV-konferensen för ett par veckor sedan. Han hade fått prover från WPI med MLV-smitta. Dessa lyckades han upptäcka i sina tester. Däremot var hela testgruppen på 50 svenska ME-sjuka ej smittade. Att hela tiden misstänkliggöra forskare som försöker bidra och med värsta möjliga konspirationsteorier känns lågt.

      Background: We wanted to search for XMRV and related viruses in Swedish ME/CFS/FM and prostate cancer patients, as well as Swedish blood donors using newly developed broadly targeted nucleic acid, virus isolation and serological techniques.
      Materials & methods: Broadly targeted real-time QPCRs: Integrase, Gag and Env PCRs for RNA and DNA were developed. They were able to detect 1-10 copies of viral nucleic acid. The PCRs were designed for variation tolerance by targeting conserved domains and the use of Megabeacon (Muradrasoli et al, 2009) probe technology. They detected murine XMRV-related ERVs but not human ERVs. Specificity was evaluated versus human, mouse and XMRV plasmid DNA. Repeatability was required for a positive result. Around 100 ng of nucleic acid was used per QPCR reaction. The amount of nucleic acid, and its amplifiability, was ascertained using a histone 3.3 QPCR (Andersson et al, 2005), and the Nanodrop spectrophotometer.

      Virus isolation: Plasma from 40 ME/CFS patients was inoculated onto LNCAP prostate cancer cells, and were tested for XMRV by QPCR after 5 days. Suspected positive cultures were passaged onto fresh cells and inoculated for another five days.
      Serology: Multiepitope serology with 22 synthetic peptides from Gag and Env of XMRV bound to colour-coded Luminex beads. Peptides were designed to react broadly by conserved sequence selection and degenerated positions.
      Patient and control samples: RNA and DNA was extracted from PBMC of 50 ME/CFS patients. The patients were selected according to the Fukuda criteria. DNA from trans-urethral resections (TURPs) from prostate tissue of 400 prostate cancer patients from Umeå University Hospital. All human samples were obtained after appropriate ethical permission.

      Results: None of the 50 ME/CFS/FM patients were positive. None of 400 prostate cancer samples were positive. None of 200 blood donor sera were positive.

      Virus isolation: Three cultures from 40 ME/CFS plasmas reacted with a few copies in the integrase PCR. One of the three could be passaged twice, but not further. The other two could not be passaged. The weakly positive LNCAP cultures had more cells floating in the medium than the negative ones. We were however not able to recover virus and viral nucleic acid from these cultures.

      Serology: Sera from 60 ME/CFS patients and 100 blood donors were tested with the multiepitope assay at 1/10. All sera were simultaneously tested with peptide-coated and noncoated bead (background). Sera which reacted over background with at least three peptides were provisionally labelled “positive”. One blood donor serum and two ME/CFS plasmas reacted in this fashion.

      Conclusions: XMRV, and putative related viruses, seem to be rare or nonexistent in Swedes, using our novel methods. The few weak and uncertain results should be further investigated

      Hei Mats😉 Jeg deiver sjelden med konspirasjonsteorier. Synser jeg, så blir dette også sagt høylytt. Grunnen for spørsmålet, var nettopp å få rese på om den svenske forsknings-gruppen hadde brukt WPI sin metodikk, noe du sier at det har de gjort, samtidig som det kommer fram i abstrakten. Når jeg lurte på om Blomberg og co har mottatt eller fått rede på om andre har tatt testen andre steder, så er vel det ett naturlig spørsmål. Uansett – det er trolig ikke Blombergs formål å referere til andre forskers funn, men å presentere sin egen forskning.

      Om en leser siste setning i hans konklusjon, så er han på langt nær ferdig med denne studien – og jeg er sikker på at det både jobbes hardt og intenst fra svenskenes side.
      «The few weak and uncertain results should be further investigated» noe som tyder på at han har funnet noe han ikke helt på daværende tidspunkt kunne verifisere, altså uklare funn.

      Vi i Norge har ikke kommet så langt med egne tester enda – vi sender disse til USA eller til Redlabs. Så all ære til svenskene som er i gang🙂

      Så Mats – vi setter virkelig pris på alle som er interessert i å belyse XMRV-saken!!!!
